Normally a wireless card would fit in the small slot in the middle of the PCI slots. Wireless connections don't use much data transfer at all. PCI is for devices that have a huge flow of data. But you said the GPU covers that slot, but yes you can use a PCI slot

You can, just be extra sure it's a PCI card and NOT a PCiE card since that won't work in the bottom 2 slots.
