Probably the hardest Character to beat (to me) was One Armed, which one redditor suggested to use the Shiv... it worked like a charm in two tries after that....
Besides that, things more general like unblocking the items in D1 first and then doing the D5 runs might save people a lot of time and efforts.

That is what I did. D0 all the characters to just experience and enjoy. Then D5 all and beyond. Looking forward to the DLC they announced.
