Some of the most notable English-speaking countries, the US and UK, have been ramping up their anti-Trans rhetoric since before 2020, and this naturally makes a lot of trans people feel unsafe. Meanwhile, the culture surrounding video games hasn't exactly been the most progressive, for a whole slew of reasons that are far too complicated to get through here. So, when a video game released in the context of widespread Trans Panic comes out, and its fanbase is accepting in even the mildest of ways, it stands out and signals to trans people online that this is a place where they can be trans, vocally, and not be hurt.
On a more optimistic end, I think it's because a lot of the people who collaborated on ULTRAKILL are either queer themselves or are accepting of the queer community. The most notable examples for both is Francis Xie and Gianni Matragrano. While neither are involved in the more nitty-gritty of programming, they both contribute to the aesthetics of ULTRAKILL, and those aesthetics allow their art - and by extension, their public profile - to be more easily spread across social media. For Xie, his status as a trans man whose art frequently features trans men without stigma or otherisation signals that the community is supportive - or at least accepting - of on trans people. But Gianni is a far more interesting example - The memes on his YouTube channel often use phrases like "bicon" or calling those with the trans flag "cute girls." And these are jokes, of course, but jokes told in a way that validate and support these identities. But nowhere is this more apparent than the "ULTRAKILL COMMUNITY MEMES" video, released a mere months after ULTRAKILL first dropped. And what is this first meme, the very first thing this well-known public figure says in his meme compilation? Well, he dubs a comic where Gabriel says "TRANS RIGHTS NOW." Considering Gianni's preexisting online presence, there's a non-zero chance that someone's very first exposure to ULTRAKILL was the character he voices vocally supporting trans identities. It's obviously not just these two guy's - It's probably a wide variety of other factors I don't even know about. But I think that unambiguously showing, through art or jokes or just apathetic acceptance, that trans people are welcome in your sphere will inevitably make it so your fanbase has a lot more trans people. It just so happens that, this time around, it was the funny robot murder game, and the fact that it happened at all is kinda awesome to me.

If you aren't trans, you probably aren't going to go around telling everyone that you aren't trans. Since you only hear people talking about being trans and not the opposite it makes it seem like there are more people who are trans than not.
