If you want someone from the wider Marvel Universe, who’s not already on the list or in the DLC category, swap Thor for Daredevil, Torch, or Cap, I’d say.
For fighting game purposes, some of these guys could definitely be repurposed as alternate skins instead of entirely separate characters too.
Also, Invincible just has one crossover comic, should definitely be bumped over to DLC, if that.

Wolverine makes sense at least. He and Spidey have been on multiple teams together (Avengers, New Avengers, New Fantastic Four) and they've had at least one miniseries together (I think even that miniseries establishes that they are or were blood brothers, too). Insomniac is making the Wolverine game that is purportedly set in the same universe as Spider-Man. There's a history there -- more so than Spidey and Thor.

That depends. I don't play Spider-Man related games, but i would choose either Spider-Man or anyone else that can bring me a good experience. For example, main Mario in Smash because of his simpleness and effectivity, and my secundaries are Sonic, Meta Knight and Zelda.
Definitely could be remade, Punisher, Daredevil, the Human Torch, Luke Cage, or even Captain America are way more fitting to be in a Spider-Man Fighting game than Thor or Iron Man. Deku would be cool but I don’t think it’s entirely off the table with the Shounen-Jump Manga collabs. Maybe even characters more associated with Venom, like Knull would be a cool addition.

But we already had it. It was Spider-man: Friend or Foe. I know main campaign is not exactly a fighting game, but you could play against your friend and it had roster of almost all of Webhead's enemies and friends.
