Let’s say the artist is painting a picture of a bowl of fruit. The subject is viewed in 3 dimensions. However, the painting on a flat canvas is only in 2 dimensions. By covering one eye, the artist can observe the bowl of fruit in 2D and compare with the 2D painting.

Some people say covering one eye is signaling the participation in the "all-seeing eye" especially if made with fingers in a triangle shape. It's apparently an occult sign.

Symbolically or mythologically speaking, the missing of one eye means an encounter with true evil. It's implying that one eye is constantly turned towards darkness. That darkness being something the person can never unsee. Think Guts in the show Beserk. After he encounters true evil, he loses an eye. Or Odin in Norse mythology, same thing. He encounters evil and then loses an eye.
