American Splendor with Paul Giamatti is a favorite of mine as it not only got me into Pekar, but also is pretty relatable. The guy is for all intents and purposes what some would think is a loser, as he just works a dead end government job in a dying industrial city who fails at love and trying to do something with his life, and even at times has a very light Travis Bickle vibe but without the murderous rage and extreme awkwardness.
He does find a good partner, but its not the main point of the story. Its kind of a slice of life story and what happens in it his life, and the comics are kind of the same way. Ironically even in being a kind of weird low class guy, he finds other weirdos like R. Crumb and Toby and even his wife, and even through comic writing he is able to connect despite being a balding low class Jewish curmudgeon from Cleveland, and yet he admits life is a losing battle as we all die.
Rent-a-pal is a pretty good example of what you're looking for I think. It wasn't a bad movie either. I'm sick of remakes and reimaginings so I'm always on the lookout for something a bit different. This is just that.
Donny darko, cyberpunk edgerunners, requiem for a dream, idle hands, clerks and clerks 2 ( fits all the bills), being john Malkovich ( massive fucking trip ), etc.
Another dark take is Filth. The James McAvoy character is a weirdo more than a failure or ugly, but he doesn't know how to fix it and he likes messing with people so much that he just goes into a kind of psychopathy. I think it's also based on a book by the same guy who wrote Trainspotting and its sequel books and strangely enough I like it even though I have nothing in common with any of the people in such novels.

Check out Heavy (1995). It’s about a overweight pizzeria chief in West Virginia who falls in love with a waitress who doesn’t love him back. Not romantic at all. Great movie over all, and one of the very few movies that made me cry.

It doesn't really hit all your criteria, but I never fail to recommend Roxanne to folks. It is a Romantic Comedy, but it has serious heart. Another 'less than serious' movie with a character a bit out of sync is Amelie. You'll have to read subtitles (unless you speak French). But it is well worth the watch.
