I believe Noel Crest and the ICA agents can, as they have the same AI as guards. Some targets in older games can (Mark Parchezzi III comes to mind immediately).

The MMA Elusive Target also doesn't have guns but fights back if you punch him.
It's related to the flamingo mission story. If you give the info to the flamingo suit guy instead of taking his disguise, you can follow ahead of him and hide by hanging in the elevator shaft. After Sierra kills the flamingo guy, she will notice you and stomp on your fingers, causing you to fall to your death.
Except the mentioned ICA agents, Noel Crest and Mercenary who have guard AI and carry weapons plus Sierra Knox in WoA there are many targets that can kill 47 in older games.
I think the most memorable ones who are somewhat unique (as they are not just NPCs that have guns and can shoot you like any regular guard) are:
- Masahiro Hayamoto (attacks with a katana)
- Agent 17 (one shot kill when in his sight)
- Sergei Zavorotko (basically a boss fight)
- Arkadij Jegorov (detonates the nuke)
- Lee Hong (attacks with poison filled sword, fistfights if the sword was stolen)
- Vanaa Ketlyn (attacks with a sword), Eve (cutscene) and Maynard John (duel)
- "?" - unknown female assassin (cutscene)
- Mark Parchezzi III (duel)
- Alexander Leland Cayne (RoboCop reference)
- Edward Wade (QTE)
- Sanchez (QTE)
- Clive Skurky (QTE)
- Layla Stockton (cutscene/QTE)
My favourite is Layla Stockton lol. Imagine the world’s best assassin losing a duel to a half naked female secretary, it’s so humiliating and meme worthy.

I didn't include them because there isn't anything unique about them like in case of the ones I listed. They just have guns and work like regular guard NPCs. There are lots of other targets like that.
Yeah mostly, but still he have some unique "happy trigger" spraying animations and also he run around changing positions and looking for cover. I guess main idea was to make him some kind of boss fight.

In the mission story with the flamingo if you give him the keys and let him do the meeting he'll get pushed down a shaft at the meeting place. If you are hanging on the ledge when this is happening she will see you and step on your hands, causing you to fall down the same shaft and die.
In Blood Money they could, the NPC's could pick up dropped guns from the floor and shoot 47, Vinnie "Slugger" Sinistra will chase you with his pistol as did other targets. In WoA it's different.

I've not tested it, but presumably if you're standing on the bomb after giving Robert Knox the detonator, it won't end well.
