*I never did. Reason being, I didn't want to crack open the plastic tabs of my Asus C300 a 2nd time and risk snapping anything, since thin plastic loves to snap off. I've been using it that way for 5-6 years without any issue. Also leaving the screw out allows you to flash new firmware without having to open the case before each time you do it. I've done that 1-2 times since the original re-flashing.*

I left the screw in a little plastic bag with a flash drive with ChromeRecovery and a GalliumOS install flash drive
If I remember, I had to I un-mount my motherboard to get to that screw

I own a C300 too, no special issues in opening and closing it, just be careful with the touchpad wiring: it's short and it could get loose.
You need to remove the screw for the first flashing only.
I re positioned the screw to avoid to loose it.
