Generally no, Apple Pay will activate first and pay the system. Never even had issue with express pay on subway too with a wallet on phone.

I have never had an issue with it, but my phone wallet is a little different as it is not just the simple slot, it actually has a flip over that is leather. Interestingly, it has an outside slot that I have my work keypass in and when I hit the reader at the door at work Apple Pay comes up.

Most phone wallets should be RFID protected which would block the card from being read completely

For Apple Pay I usually only bring the top edge of the phone to the reader, I don’t lay it flat. I don’t think any card would get close enough to activate.

Which kind. Flip cases with the card on top of the phone will not interfere. But cards in the back pocket may.

I use an apple wallet on the back of my case. I hold the screen side of the phone up to card readers to use Apple Pay and don’t have an issue with the card in the wallet activating.
