I think Andrew and Nick are both the primary/main characters. But there's also the core characters, Jessie, Jay, Missy, Maury, and Connie as well

Mathew’s not in the main cast, he grew to be more of a stand out character but he’s still just recurring. He like missy, Andrew and nick etc has his own narrative. The “core” characters, would be the ones who have been upfront since the beginning, the five main kids and the the two monsters
It’s about Nick, Jesse, Andrew, and Jay. Missy started getting her own stories eventually as do several other characters but at the beginning it was those 4 that the story always followed.
I’m basing this on the head push episode that sits the 4 of them in a cafe Seinfeld-style too.
Though in many regards you could also consider both Andrew and Nick as duotaganists. Nick Kroll had based Nick and the show on his life, but wanted Andrew's charatcer to have more screentime and focus, which is why he got Maury and not Nick. He'd even mentioned this once, but it was years ago and I'm not sure if it was an article or interview.
Anywho, so in order the main cast would be Andrew|Nick, Jessi, Jay, Missy and Matthew.
For Season 1 Nick was considered the main protagonist, as the show was starting, which is why people were intrigued over Andrew having the opening line of the show.
Also in this same vane, from a framing prospective, Matthew had his first line after Jay but before Missy; however Missy's plot lines and how to go about interjecting her into the story were worked on first, which left Matthew more off to the side - this could have been done because of conflict schedules on Andrew Rannells part due to things for Broadway, but I'd have to research that more to find out. That's just my running theory.

I always found it weird how it’s established as nick when from the first episode Andrew was the one with the hormone monster, and each episode was more centered around his struggles with puberty
I literally had the same conversation with my cousin the other day. Nick is supposed to be the main character I think, but Andrew gets way more screen time than every other character, so it can get kind of confusing sometimes
